How the HTPTM process works:
Phase I: Immunization (Day 1)
- Three HTPTM mice are immunized. Your immunogen is emulsified with Freund’s Complete Adjuvant. Booster injections are emulsified with Freund’s Incomplete Adjuvant. After the last immunization, the antibody titers are assessed by ELISA.
- Immunogen requirements: 1.5 to 3 mg suitable for immunizations and ELISA plate coating. 0.5 mg negative control protein. Let us know if this quantity is difficult to obtain; we can work with smaller amounts of antigen. If the protein is not available, we can design and synthesize peptides for immunization and screening.
Phase II: Fusion (Day 21)
- Cells from immunized mice are prepared and fused with myeloma cells. Viable hybridomas will be HAT selected and screened for antigen-specific antibodies by ELISA.
Phase III: Screening (Finished by Day 35-45)
- Screening for most targets is done via ELISA. We also have the capability to screen using flow cytometry, western blotting, affinity ranking, and others upon request.
- Up to 48 antibody secreting hybridomas per fusion with the best characteristics will be grown up and re-tested.
- Optional: Supernatants from positive hybridomas can be shipped to you for evaluation. This step usually takes place 40 days after immunization.
- Note: If the customer decides to screen the supernatant samples, we ask that you are prepared ahead of time to receive and assay them so that we may move forward in a timely manner.
Phase IV: Subcloning (Day 45-54)
- Up to five (5) positive parental hybridomas are subcloned. One (1) vial of each parental hybridoma is frozen down with about 2x106 cells/vial for long term storage. Single colony wells are screened for antibody secretion by ELISA.
- Two (2) subclones from each parental hybridoma are expanded and frozen down into three (3) vials each.
Deliverables
Day 40:
- Up to 48 supernatant samples for our customer to screen for selection of hybridomas with desired characteristics for subcloning and delivery
Day 60:
- Up to 10 hybridoma subclones from 5 parentals specific to antigen by ELISA are cryopreserved in two vials per subclone.
- Supernatant samples from each subclone
- Isotype of each subclone
- All subclones and parentals (1 vial each) are stored in our cell bank for 6 months without additional charge.
- Report detailing the work performed by Abpro

Guarantee
We guarantee that the hybridoma clones supplied to customers will be immunogen positive by ELISA. If we cannot deliver at least one clone producing specific antibody, you will not be charged for the full cost of the project. We have a bill-by-phase payment structure which is designed to protect our customers from the inherent risk in hybridoma development.
